Differentiated cells can be reprogrammed to a state such as embryonic by transferring nuclear content to oocytes or with fusion with embryonic stem cells. Little is known about the factors that induce this reprogramming. Here, we show induction of pluripotent stem cells from embryo mouse or adult fibroblasts by introducing four factors, Oct3 / 4, Sox2, C-MYC, and KLF4, under the condition of the culture of ice cells. Unexpectedly, nanog can be discarded.

These cells, which we appoint IPS cells (induction of pluripotent stem), show morphology and the growth properties of ice cells and express ice marker genes. The subcutaneous transplant of IPS cells to naked mice resulted in tumors that contain various networks from the three layers of germs.

After injection becomes a blastocist, IPS cells contribute to the embryonic development of the mouse. This data shows that pluripotent stem cells can be directly generated from fibroblast culture by adding only a few factors specified.

Map and measure mammal transcriptom by RNA-SEQ.

We have mapped and measured the mouse transcriptom by sorting them and recording how often each gene is represented in the sequence sample (RNA-SEQ). This provides a digital size of the existence and prevalence of transcripts of known and previously unknown genes. We report reference measurements consisting of 41-52 million mapped 25-base-pair reap for poly (a) – RNA levels from adult mouse brains, muscle tissue and frame.

We use RNA standards to measure the prevalence of transcripts and to test the linear range of transcript detection, which stretches five magnitude. Although> 90% of the reading mapped uniquely falling in known exon, the remaining data shows a new and revised model model, including changed or additional promoter changes, exon and 3 ‘non-translated areas, and new candidate microorda precursors.

The RNA Splice event, which is not easily measured by the expression of the microarray standard gene or serial analysis of the gene expression method, is detected directly by mapping the sequence of junction connections. We observed 1.45 x 10 (5) different connections, and alternative alternative splices, with 3,500 different genes expressing one or more alternative internal connections.

Order of the human genome.

Sequence Consensus Base 2.91 billion (BP) from the Euchromatic section of the human genome is produced by the Shotgun sequencing method of all genomes. The sequence of DNA 14.8 billion BP was produced for 9 months from 27,271,853 high-quality order reading (5,11 times the genome) of the two ends of plasmid clones made from DNA from five individuals.

Two assembly strategies – assembly of all genomes and regional chromosome assembly – used, each incorporates data order from the Celera and a public-funded genome effort. Public data is embraced to the 550-BP segment to make 2.9 times coverage of the sorted genome area, without entering the bias attached to the cloning and assembly procedures used by publicly funded groups.

This brings effective coverage to assemblies up to eight times, reducing the number and size of the gap in the final assembly of what will be obtained with the coverage of 511 times. Two assembly strategies produce very similar results that mostly agree with independent mapping data. Assemblies effectively cover the Euchromatic region of human chromosomes.

More than 90% of the genome in seizure scaffolding 100,000 BP or more, and 25% of the genome in the scaffolding of 10 million BP or greater. Analysis of the order of the genome revealed 26,588 protein coding transcripts that have strong strong evidence and an additional of around 12,000 genes that are reduced computing with mouse compatibility or other weak supporting evidence.

Even though the cluster solid genes are clear, almost half the gene is dispersed in a low G + C sequence separated by a large channel from the seemingly unbodged. Only 1.1% of the genome recommended by the exon, while 24% in introns, with 75% of the genomes become intergenic DNA. Duplication of Segmental Blocks, range in size to chromosome length, abundant throughout the genome and reveal the history of complex evolution.

Random pairs of human haploid genomes are different at the level 1 of BP per 1250 on average, but there are heterogeneity marked at the level of polymorphism throughout the genome. Less than 1% of all SNP results in variations in protein, but the task determines which SNP which has functional consequences remains an open challenge.
